Molecular Mechanisms and Therapeutic Approaches to the Treatment of Metabolic Associated Fatty Liver Disease (Meta-MAFLD)-PRIN PNRR 2022-MUR

The "Molecular Mechanisms and Therapeutic Approaches to the Treatment of Metabolic Associated Fatty Liver Disease (Meta-MAFLD)" project aims at exploiting the molecular mechanisms characterizing the metabolic associated fatty liver disease (MAFLD) and at discovering new possible natural products which can contribute to the treatment of this disease. Some evidence has suggested that dysregulation of tyrosine phosphatase type 1 (PTP1B) and the nuclear receptor peroxisome proliferator-activated receptor ? (PPAR?) may be involved in the pathogenesis of NAFLD/MAFLD and the modulation of these molecules may improve this metabolic disfunction. In this project, we have selected two structural classes of both natural and natural-inspired compounds: i) the plant derived isoprenoid-substituted benzoic acid derivatives amorfrutins and amorfrutin-type phytocannabinoids; ii) the synthetic and semi-synthetic phosphorylated polyketides designed on the model of marine metabolite phosphoeleganin. Sustainable organic semi-syntheses or simple and easily scalable total syntheses will be adopted to produce chemical analogues/focused libraries within these classes of compounds. This choice is based on their promising effects previously evidenced as ligands of PTP1B and PPAR?. Moreover, for some of these compounds, the ability to improve the insulin signaling pathway has been already evaluated in other insulin-sensitive cell types. Thus, the specific objectives of Meta-MAFLD project are: 1. Understanding how the phosphoeleganin derived/inspired and/or amorfrutins-based compounds may improve insulin sensitivity and inflammatory response in liver and skeletal muscle cells. 2. Understanding whether and which cytokines, chemokines, hepatokines, and myokines may be regulated by the phosphoeleganin derived/inspired and/or amorfrutins-based compounds 3. Assessing the effects of hepatokines and myokines in the crosstalk of liver/skeletal muscle with each other as well as with the adipose tissue.

Sistema Immunitario Comportamentale e Conformità sociale: aspetti psicometrici.

l framework teorico del Sistema Immunitario Comportamentale (SIC) sostiene che abbiamo sviluppato processi cognitivi, reazioni affettive e comportamenti per evitare minacce da patogeni. Nel SIC, l'emozione centrale del disgusto svolge un ruolo chiave, scatenando reazioni di evitamento appropriate per prevenire il contagio. Nonostante l'importanza di adottare comportamenti appropriati per evitare patogeni, molte sfaccettature del SIC sono ancora in gran parte sconosciute. La conformità sociale (CS) potrebbe rappresentare una risposta adattativa di fronte a una minaccia invisibile da patogeni. Vi è una robusta evidenza che i livelli individuali di tratti correlati al BIS sono associati a diverse misure di CS. Tuttavia, manca ancora un chiaro supporto per un legame causale tra l'attivazione del BIS e successivi cambiamenti nella CS. Una possibile ragione per tale discrepanza è che le attuali misure delle differenze individuali nel BIS potrebbero non catturare validamente il costrutto sottostante. Ad esempio, queste misure potrebbero riflettere bias di risposta condizionati da diversi sottogruppi di popolazione. Il primo obiettivo del progetto Behavioral Immune System and Social Conformity (BISSCo) è investigare questa possibilità. Uno degli aspetti critici della nostra reazione a minacce invisibili come i patogeni è prestare maggiore attenzione alle altre persone e conformarsi infine al comportamento della maggioranza, poiché può essere una strategia efficace per evitare il rischio di infezione. Di conseguenza, anziché promuovere una CS indiscriminata, il BIS dovrebbe agire aumentando l'attenzione selettiva verso coloro che percepiamo come condividere le stesse norme culturali e pratiche. Se così fosse, ci si potrebbe aspettare un aumento della conformità, specialmente verso gruppi di riferimento salienti (ad esempio, gruppi interni) in contesti ecologicamente rilevanti (ad esempio, norme alimentari). Il secondo obiettivo di BISSCo mira a esplorare come l'identità di gruppo e la percezione delle norme sociali influenzino il legame tra conformità e BIS.

Exploring the role of SGLT-1 driven intestinal glucose absorption and altered intestinal mucosa integrity in the pathogenesis of non-alcoholic fatty liver disease

The pathophysiology underlying non-alcoholic fatty liver disease (NAFLD) is not completely clarified. We have recently reported that subjects with ultrasound-defined NAFLD have increased duodenal of levels the glucose carrier sodium/glucose co-transporter 1 (SGLT-1) and early postprandial hyperglycemia. Inhibition of intestinal SGLT-1 has been shown to prevent high fat diet induced hepatic steatosis, and gut epithelial barrier damage. An altered gut barrier integrity is implicated in NAFLD pathogenesis. Notably, preclinical evidence suggests that high glucose may impair intestinal mucosa integrity. Activation of endoplasmic reticulum (ER) stress plays an important role in cellular damage associated with glucotoxicity, and may affect intestinal barrier integrity. Based on these evidence, it is plausible that augmented duodenal SGLT-1 abundance results in a higher transport of dietary glucose from the intestinal lumen into enterocytes, activating ER stress which may affect duodenal mucosa integrity, thus contributing to NAFLD development and progression. This proposal seeks: 1) to evaluate whether subjects with hepatic steatosis and more importantly those with NAFLD-related hepatic fibrosis assessed by transient elastography, display increased duodenal dietary glucose absorption driven by augmented duodenal levels of SGLT-1, along with ER stress activation and impaired mucosal defense; 2) to examine the role of ER stress in high glucose induced duodenal barrier dysfunction; 3) to test whether SGLT-1 inhibition may counteract the deleterious effects of hyperglycemia on intestinal ER homeostasis and mucosal integrity in subjects with NAFLD. The expected findings of this proposal will demonstrate the link between NAFLD and its severity and an upregulation of duodenal SGLT-1 mediated glucose absorption/ER stress/mucosal permeability axis, and will support application of strategies targeting the up-regulated SGLT-1 driven glucose absorption in the treatment of NAFLD.
